Molecular characterisation and similarity relationships among iranian basil (Ocimum basilicum L.) accessions using inter simple sequence repeat markers Rev. Ciênc. Agron.

The study of genetic relationships is a prerequisite for plant breeding activities as well as for conservation of genetic resources. In the present study, genetic diversity among 50 Iranian basil (Ocimum basilicum L.) accessions was determined using inter simple sequence repeat (ISSR) markers. Thirty-eight alleles were generated at 12 ISSR loci. The number of alleles per locus ranged from 1 to 5 with an average of 3.17. The maximum number of alleles was observed at the A7, 818, 825 and 849 loci, and their size ranged from 300 to 2500 bp. A similarity matrix based on Jaccard's coefficient for all 50 basil accessions gave values from 1.00-0.60. Assessment of genetic diversity among Pakistani wheat (Triticum aestivum L.) advanced breeding lines using RAPD and SDS-PAGE Electron. J. Biotechnol.

Genetic diversity was assessed among 32 advanced wheat breeding lines included in the National Uniform Wheat Yield Trials (2006-07) of Pakistan using molecular (DNA) and biochemical (SDS-PAGE) markers. Of the 72 RAPD primers used for initial screening, 15 were found polymorphic. A total of 140 bands (61.4% polymorphic) were generated by the 15 random decamer primers. Genetic similarity coefficients ranged from 0.81 to 0.94 for rainfed and from 0.70 to 0.93 for the normal seeding date group. Cluster analysis using the unweighted pair group method of arithmetic averages (UPGMA) clustered the 32 advanced wheat breeding lines into one major and three small groups. The solitary egg parasitoids (Hymenoptera: Scelionidae) of Pentatomidae species complex are potential candidates for soybean (Glycine max L.) integrated pest management programs in Brazil. The correct identification of species and strains of these parasitoids is necessary to implement the use of biological control. Polymerase chain reaction (PCR) was used to generate random amplification of polymorphic DNA (RAPD) from male individuals of Telenomus podisi Ashmead and Trissolcus spp. collected in a soybean field in Brasília, Brazil in 1996. The genetic diversity of a single nucleotide polymorphism (SNP) at the exon 20 (T945M) of the leptin receptor gene (LEPR) and of three short tandem repeats (STRs BM7225, BMS694, and BMS2145) linked to LEPR was investigated in three beef cattle herds (Brangus Ibagé, Charolais, and Aberdeen Angus). A cheap and effective new method to analyze the T945M polymorphism in cattle populations was developed and the possible role of these polymorphisms in reproduction and weight gain of postpartum cows was evaluated. In order to extend the genetic data on the Sciaenidae fish family, the present study had the purpose to characterize PCR-generated 5S rDNA repeats of twelve species of this group through PAGE (Polyacrylamide Gel Electrophoresis) analysis. The results showed the occurrence of at least two different 5S rDNA size classes in all the species. Moreover, 5S rDNA repeats of one of the studied species - Isopisthus parvipinnis - were cloned and subjected to nucleotide sequencing and Southern blot membrane hybridization analyses, which permitted to confirm the existence of two major 5S rDNA classes. O propósito deste trabalho foi determinar a similaridade genética entre acessos de germoplasma e cultivares comerciais de três espécies de Brachiaria (inter e intraespecífica), por meio de marcadores RAPD. The aim of this work was to evaluate the carotenoid content and genetic variability of banana accessions from the Musa germplasm collection held at Embrapa Cassava and Tropical Fruits, Brazil. Forty-two samples were analyzed, including 21 diploids, 19 triploids and two tetraploids. The carotenoid content was analyzed spectrophotometrically and genetic variability was estimated using 653 DArT markers. The average carotenoid content was 4.73 µg.g-1, and ranged from 1.06 µg.g-1 for the triploid Nanica (Cavendish group) to 19.24 µg.g-1 for the triploid Saney. The tropical multipurpose shrub legume Cratylia argentea is well adapted to acid soils of low to medium fertility and has excellent drought-tolerance. The present study aimed to estimate and characterize the genetic divergence between determinate and indeterminate snap bean accessions from the Universidade Estadual de Londrina (UEL) germplasm bank based on amplified fragment length polymorphism (AFLP) markers. A total of 40 and 32 accessions with determinate and indeterminate growth habits, respectively, were characterized for this purpose. Polyembryony and identification of Volkamerian lemon zygotic and nucellar seedlings using RAPD PAB

The objectives of this work were to evaluate the frequency of polyembryony, and to identify zygotic and nucellar seedlings of Citrus volkameriana using RAPD. Twenty-five polyembryonic and eight monoembryonic seeds were cultivated in vitrofor six months. DNA from seedlings was extracted and used in combination with five RAPD primers to identify zygotic or nucellar origin of the seedlings. Environmental conditions of the year affected significantly (P<0.05) the morphological characteristics of fruitsand the number ofembryos per seed. Development and characterization of fifteen polymorphic microsatellite loci in Bryconamericus aff. iheringii (Teleostei: Characidae) and cross-amplification in related Characidae species Neotropical Ichthyology

ABSTRACT Data on 15 novel microsatellite loci from the Neotropical fish Bryconamericus aff. iheringii are presented here. Analyses of 32 individuals from four different streams revealed 192 different alleles, ranging from four to 32 alleles per locus (mean of 12.8 per locus). Observed and expected heterozygosities ranged from 0.094 to 0.813 and 0.205 to 0.952, respectively. These loci showed high polymorphic information content and will be a resource for genetic studies of B. aff. iheringii. Furthermore, several loci also amplified other small Neotropical Characidae (Piabarchus stramineus and Piabina argentea) and should be useful for these species.

A polymerase chain reaction (PCR)-based approach was employed to isolate putative alleles of the chromoplast-specific lycopene beta-cyclase (CYCB) gene from wild and cultivated tomatoes [Solanum (Section Lycopersicon)]. The objective of this work was to establish an effective PCR protocol by testing DNA samples from distinct germplasm accessions with a primer pair designed to selectively target conserved regions present in the available CYCB sequences. The gene Pi-ar confers resistance to Pyricularia grisea race IB-45 in a somaclone derived from immature panicles of the susceptible rice (Oryza sativa) cultivar Araguaia. RAPD technique was used to identify molecular markers linked to this gene utilizing bulked segregant analysis.  GenomicLand is free software intended for prediction and genomic association studies based on the R software. This computational tool has an intuitive interface and supports large genomic databases, without requiring the user to use the command line. GenomicLand is available in English, can be downloaded from the Internet (https://licaeufv.wordpress.com/), and requires the Windows or Linux operating system. The software includes statistical procedures based on mixed models, Bayesian inference, dimensionality reduction and artificial intelligence. Examples of data files that can be processed by GenomicLand are available. The examples are useful to learn about the operation of the modules and statistical procedures.

The software includes statistical procedures based on mixed models, Bayesian inference, dimensionality reduction and artificial intelligence. Examples of data files that can be processed by GenomicLand are available. The examples are useful to learn about the operation of the modules and statistical procedures.

HSF1 and HSPA6 as functional candidate genes associated with heat tolerance in Angus cattle R. Bras. Zootec.

ABSTRACT The purpose of this study was to access and characterize single nucleotide polymorphisms (SNP) located within the HSF1 and HSPA6 candidate genes for adaptability in Angus breed raised in subtropical climate. Samples of DNA from 20 animals representing extreme phenotypes for adaptability traits were obtained. Sequence variations in the candidate genes were described by sequencing target regions. We identified 12 SNP located in the HSF1 gene. Moreover, four of the six SNP found in the HSPA6 gene cause amino acid substitutions in protein-coding regions. We also identified a representative SNP (called tag SNP) in a region of the HSF1 gene with high linkage disequilibrium (r2 = 0.87) that may represent 11 SNP located in this gene. Minor allele...
